Examples of cryoultramicrotomy.

Cryoultramicrotomy utilizing a Leica UCT cryoultramicrotome for optimizing epitope availability and morphological preservation for immunogold labeling.  

The examples below are rat anterior pituitary GH3 cells immunolabeled with antibody against an enzyme specific to Golgi with 10 nm gold. 

Note the clean stacks of Golgi with precise localization of 10 nm gold antibody.  Click here for a 3D reconstruction of Golgi in the same cell type from data collected with the old BioRad MRC 600 confocal microscope.
